제목: 합성생물학 기반 미생물 세포공장 개발

 

내용: Engineering of microbial cells related to health, food, and chemical industry is growing. However, it remains many challenges, which are associated with the complex regulatory networks underlying the metabolism and evolutionary adaptation of microbial cells. In order to tackle these challenges and enable effective engineering of microbial cell factories, we have adopted the Design-Build-Test-Learn paradigm, largely facilitated by the methods development within synthetic biology. Here, we show some examples; (1) enhancement of our understanding of 3'-UTR regulatory functions and a diverse collection of tunable 3'-UTR bioparts with a wide termination strength range, (2) engineering of Bacteriodes thetaiotaomicron for heterologous production of non-native butyrate at therapeutically relevant concentrations in the human gut using its expanded genome-scale metabolic model (GEM), (3) evolutionary and metabolic engineering in tandem to construct an Escherichia coli strain with improved GABA production using glycerol as the feedstock carbon, and (4) engineering of Eubacterium limosum for C1 gas feedstokcs utilization. These studies highlight that microbial cells can serve as effective means for live microbial therapeutics and value-added commodity chemical production to meet the demands of biosustainability.
